彩叶草羟基丙酮酸还原酶基因CbHPR的克隆及分析   总被引:1,自引:1,他引:0  
为研究C3植物彩叶草的光呼吸作用,根据获得的HPR EST片段,采用RACE和文库结合的方法,克隆了HPR蛋白的全长cDNA,命名为CbHPR(GenBank accession No.EF125078).序列分析结果表明,该cDNA全长为1 393 bp,包含一个1 161 bp的ORF框,编码386个氨基酸;其5′-UTR区含有2个终止子TGA,3′-UTR区具有推测的加尾信号AATAAA;CbHPR蛋白C端具定位于微体的转运信号S-K-L,具有1个保守的2-Hacid_dh_C domain结构域(D-异构体-羟基酸脱氢酶-NAD结合结构域)、5个S磷酸化位点、4个T磷酸化位点和6个Y磷酸化位点.PSORT定位分析显示,CbHPR定位于叶的过氧化物酶体中.多序列比较和进化树分析表明,CbHPR与其他植物HPR一致性高达87%~90%,与拟南芥AtHPR具有相似的功能.CbHPR基因的克隆与分析,为进一步研究该基因在彩叶草的光呼吸作用中的表达调控奠定了基础.  相似文献

Quantitative determinations by gas chromatography-mass spectrometry ofindole-3-acetic acid (IAA) and abscisic acid (ABA) in growing leaves ofColeusblumei plants show parallel declines in leaf concentrations of bothhormones,except in leaf number 3 (about three-fourths of full size) where IAA level wasthe lowest of those measured. Expansion of the most recently unfurled leaf tofull size serves, in effect, to dilute both IAA and ABA about 1.7 to 1.Althoughabsolute levels of leaf IAA varied as much as an order of magnitude from onebatch of plants to another, ABA levels were proportional to the IAA level withan overall correlation coefficient of 0.91. Evidence, both correlative andcausal, for the determination of ABA status by IAA—and of IAA status byABA—in leaves and other developing organs is summarized.  相似文献

为了解乙二胺二琥珀酸(EDDS)诱导植物耐受硒(Se)胁迫的生理机制, 以彩叶草(Coleus blumei)为材料,采用营养液培养的方法、借助傅里叶变换-衰减全反射红外光谱(FTIR-ATR)和扫描电子显微镜-X-射线能量色散谱(SEM-EDXS)分析方法及生理指标的变化,研究1.0 mg/L Se胁迫条件下添加0、0.5、1.0、1.5、2.5、5.0 mmol/L EDDS 对彩叶草根系化学成分变化的影响。利用FTIR-ATR图谱分析发现,随着EDDS处理浓度的提高,彩叶草根系透射峰所对应峰形基本不变,而参与Se吸附的基团如羟基、酰胺基和指纹区等的透射峰发生了不同程度的位移。FTIR-ATR的特征峰与彩叶草根系响应Se胁迫的各生理指标变化趋势基本一致,且FTIR-ATR比传统的生理指标测定更敏感、便捷。SEM-EDXS扫描还发现随着EDDS处理浓度的升高,根系中K、Mg、Fe、Si 等元素的含量升高,而营养元素Ca含量降低。该研究结果可为深入了解EDDS处理下彩叶草对Se胁迫的响应机理提供科学依据。  相似文献

A high frequency shoot organogenesis and plant establishment protocol has been developed for Coleus forskohlii from leaf derived callus. Optimal callus was developed from mature leaves on Murashige and Skoog (MS) medium supplemented with 2.4 μM kinetin alone. Shoots were regenerated from the callus on MS medium supplemented with 4.6 μM kinetin and 0.54 μM 1-naphthalene acetic acid. The highest rate of shoot multiplication was achieved at the sixth subculture and more than 150 shoots were produced per callus clump. Regenerated shootlets were rooted spontaneously on half-strength MS medium devoid of growth regulators. The in vitro raised plants were established successfully in soil. The amount of forskolin in in vitroraised plants and wild plants was estimated and found that they produce comparable quantity of forskolin. This in vitro propagation protocol should be useful for conservation as well as mass propagation of this plant. Summary A protocol has been developed that leads to the development of complete plantlets of Coleus forskohlii within 35–40 d by culturing stem tip explants in MS medium containing 0.57 μM indole-3-acetic acid and 0.46 μM kinetin through direct multiplication at the rate of 12.5 shoots per explant. About 100% shoots rooted and micropropagated plants were successfully established in soil after hardening with a high survival rate. The significance of the present micropropagation protocol of C. forskohlii is the formulation of growth regulators which effected very fast multiplication of the plant (time reduced to one-third of the hitherto known methods).  相似文献

bstract Suspension cultures of Coleus blumei (Lamiaceae) treated with either an elicitor preparation from the culture medium of the phytopathogenic oomycete Pythium aphanidermatum or with methyl jasmonate enhanced accumulation of rosmarinic acid approximately threefold. The specific activities of phenylalanine ammonia lyase and rosmarinic acid synthase were also enhanced after addition of the fungal elicitor. The addition of methyl jasmonate transiently increased activities of phenylalanine ammonia lyase and hydroxyphenylpyruvate reductase, whereas the activity of rosmarinic acid synthase was not stimulated and the activity of tyrosine aminotransferase was slightly and constantly enhanced. Methyl jasmonate stimulated rosmarinic acid accumulation not only when added directly to the culture medium, but also when it could reach the cells only via the gas phase. Many doubts still persist even today when it comes to selection of the solvents for extracting the active constituents from various Indian medicinal plants. This study was aimed at assessing and establishing the best solvent for extracting the active constituents from 10 plant extracts. Thin layer chromatography (TLC) was used to separate and establish the active constituents present in each of the medicinal plants. Active constituents from each plant were extracted by using three different solvent systems namely diethyl ether, chloroform and hexane and were tested against three species of gram negative and three species of gram positive bacteria (Escherichia coli, Pseudomonas aureginosa, Streptococcus pneumoniae, Aeromonas hydrophila, Staphylococcus aereus, Bacillus cereus) by means of agar well diffusion assay. Studies on the antioxidant activity studies were also carried out for these plant extracts by using Diphenylpicryl-hydrazyl (DPPH) method. For the antimicrobial activity, the study revealed that among the selected plants, Azadiracta indica, Pongamia pinnata, Aloe barbadensis had the maximum antibacterial activity. Among the extraction procedures diethyl ether was found to be the best solvent that could be used for the extraction procedure. On the antioxidant activity part, Coleus amboinicus and Calotropis procera were found to have high antioxidant activity of 91.64% and 88.72% respectively and the further results are reported and discussed.  相似文献

本文报导了,从产于滇东北的毛喉鞘蕊花(Coleus forskohlii Brig)的根茎中分离到四个二萜化合物,经光谱分析鉴定,其结构分别为1α,6β,7α-triacetyl-coleonol—B(1),I-acetyl—forskolin(2),coleonol-B(3),和deacetyl-forskolin(4)。化合物(1)和(2)是首次从该植物中分离到。  相似文献
